Wild type AaCas12b mRNA enables the expression of an Acidicyclobacillus acidophilus-derived CRISPR-associated protein 12b (Cas12b), an RNA-guided DNA endonuclease that belongs to the type V CRISPR-Cas system. When co-transfected with a single guide RNA (sgRNA) targeting specific genomic loci, AaCas12b protein binds to the DNA through complementary base pairing with its sgRNA, inducing site-specific double-strand breaks (DSBs) at the target site. Unlike conventional SpCas9, AaCas12b exhibits stringent PAM recognition specificity (TTN motifs) and demonstrates significantly reduced off-target activity due to its unique structural architecture, including an extended recognition helix and enhanced DNA: RNA hybrid stability.
